Afinitor® is an oral prescription medication used to treat certain types of advanced hormone receptor-positive, HER2-negative breast cancer in postmenopausal women. Afinitor is also used to treat advanced kidney cancer after failure of treatment with Sutent® (sunitinib) or Nexavar® (sorafenib).
Afinitor tablets contain 2.5 mg, 5 mg, 7.5 mg, or 10 mg of everolimus. The recommended adult dosage for advanced breast cancer is 10 mg once daily. For advanced kidney cancer, the usual dosage is 10 mg once daily.
Afinitor works by blocking mTOR, a protein involved in tumor growth and blood vessel development in cancer cells. This can slow or prevent advanced cancer from progressing further.
Common side effects may include mouth sores, infections, feeling weak/tired, cough/cold symptoms, diarrhea, constipation, fever, headache, swelling in arms/legs, rash, vomiting, and loss of appetite.
Do not take Afinitor if allergic to everolimus. Use caution with medications that inhibit or induce CYP3A4 enzymes. Avoid grapefruit and grapefruit juice. Tell your doctor about all medications and medical conditions.
